Cancer Metabolomics and Proteomics

Mass spectrometry (MS) is the premier technology for detecting and quantifying a wide range of biomolecules – enabling survey analyses like metabolomics, lipidomics, and proteomics. The UW Carbone (UWCCC) Cancer Metabolomics and Proteomics (CAMP) shared resource serves investigators at UWCCC through consultation, education, and MS-expert match-making to enable high quality MS-based analyses for cancer research. The CAMP resource builds upon the unique campus MS infrastructure and leverages it to provide unparalleled MS technical capability and support for UWCCC member research.

CAMP comprises a network of Expert MS laboratories and MS Cores. CAMP serves as a resource and access point to cutting-edge mass spectrometry technologies with a goal of 1) educating, 2) advising, 3) matching members to MS-experts on campus.

UWCCC members that wish to incorporate metabolomics and proteomics into their research should follow this process.

  1. Contact Dr. Coon or Dr. Overmyer to discuss project goals, research innovation and impact, and cancer relevancy.
  2. Drs. Coon and Overmyer assist UWCCC members with the match-making process with one of the CAMP MS-Expert labs.
  3. For projects needing some initial funding support, there are small microgrants available. Microgrants of are intended to jump start the collaborative projects between CAMP Expert Labs and UWCCC members. For these microgrants, project summaries will be reviewed by the CAMP scientific advisory committee and funds will be awarded based on merit and feasibility of the project.

Education and consultation

CAMP leadership meets one-on-one with UWCCC PIs to discuss project proposals, learn about the project background, overarching goals, timelines, etc. Similarly, they explain the capabilities and limitations of MS as relevant to the proposed work.

MS technology – Proteomics and PTMs

The CAMP network possesses extensive expertise and cutting-edge instrumentation to provide deep global analysis of proteins and their PTMs. Most of the MS instruments are outfitted with nano high pressure liquid chromatography (nHPLC) systems and have a variety of dissociation technologies including collisional activation, electron transfer, and photoactivation. Further, multiple are outfitted with ion mobility – either traditional or high-field asymmetric waveform ion mobility (FAIMS). Our expert labs routinely perform comparative analysis on scale of dozens to hundreds of biological samples, depending on the project.

MS technology – Metabolomics and Lipidomics

CAMP expert labs are also at the cutting-edge of metabolite and lipid analysis – routinely quantifying ~1,000 unique lipids and metabolites from complex biological samples. Typical analyses involve extracting the target compounds of interest and performing LC or GC-MS on the resultant complex mixtures of compounds. Both discovery and targeted analyses are routinely done across these various CAMP expert labs. Of note, are the labs of Amador-Noguez and Fan, who are uniquely setup to perform metabolic flux assays. And the MS imaging capability of Li is also highly relevant for spatial mapping of lipids and metabolites in various tissues and tumors.

Any published research which received funding from the UWCCC Cancer Center Support Grant (CCSG), including the use of UWCCC Shared Resources to generate or analyze data or conduct clinical trials, must reference the UWCCC. See Acknowledging UWCCC in Publications, Posters and Presentations. Please acknowledge “UWCCC Cancer Metabolomics and Proteomics Shared Resource” for any work supported by CAMP. Thank you!
